Of course Uncle Ben is going to die. That's a given with Spider-Man's origin. I'd say it's even more important than the spider bite.

I'm talking about as far as what is most important to show in a telling of the origin. I'm more interested in the why than the how. Uncle Ben's death is Spider-Man's raison d'être. Without Uncle Ben's death, Spider-Man wouldn't be a crimefighter.

If you're going to show Uncle Ben's death, you might as well show the bite. They happen within days/weeks of each other.

If I had to skip one, I'd skip the bite and see Uncle Ben's death. The truth is that there's no significance to the spider other than a source of power. It's a plot device that isn't part of a deeper allegory, nor does it touch upon themes other than possibly the randomness of fate and the unpredictability of life.
Uncle Ben's death is what leads him to being the person Peter is. I'll take character development over a plot device anyday.

If it were me making the movie I wouldn't touch the origin with a barge pole. Get on with it and anything the audience needs explained tell it in flashbacks. I think the general audience just isn't excited to see Spider-Man's origin again after only 10 years, and I can't blame them, so trying to use that as the selling point like the "teaser" trailer did was a huge mistake, in my opinion.
