Well, there hasn't been any news on the film's progression. Other than Tyrese supposedly being interested in the role, and preparing himself for it. But news such as a confirmed cast, or release date has yet to be officially set.

Doubtful. But not completely out of thought I'm sure. There could be someone to take his place, though. That is, if Diamondback is still going to be the main villain. Howard is going to be busy as James Rhodes in the Iron Man film.

I think it'll have to be. Superman VI will be 2009, and 2010 is currenly DC movie free, but that's too far off as Warner Bros are already searching for a Wonder Woman.
