MJ might not be Peter's first love, and it might even be more interesting to use Gwen in a movie, but MJ has become the classic love interest, and that is whom the public know too. She is to Peter what Lois Lane is to Clark Kent. In a Superman film (although it has been done in Smallville), it might be interesting to have Lana as the main love interest and explore her relationship with Clark, but the public are expecting to see Lois. The general public know Lois as Clark's woman, but not everyone knows Lana. To a similar extent, that is the same with Mary Jane, so there is a certain level of expectation, and something less might affect the box office returns. Comics are like the modern equivalent of Greek Myths. We expect to see Paris fall for Helen of Troy and not some other girl first, and we expect to see Achilles shot by an arrow into his heel. These reboots are just alternate retellings of the same myth, so to have Gwen as the love interest, while fine in the comics, might not fly so well with the movie-going public.
