Trying to make a Joker in the mold of Nolan's movies I think we could be shown some of the spirit of the character in the following ways. Language will be most important. Joker's word choices are great. Most of the time he has a brilliant vocabulary with a very enthusiastic and purposeful delivery. Particularly eloquent and measured during sadistic moments, before the punchline is delivered. A few phrases put together somewhat poetically goes a long way to show the wit. Then, to bring forward his mentality, a few instances where he absentmindedly breaks from the show (like the "oh right, dead" line). Kind of informs us that he sees himself as giving a performance a lot of the time and in those quick moments of lucidity he forgets to continue the performance. Almost like sliding in and out of character. Brings that unpredictability too. At one point maybe becoming furious, like a director, at someone he shoots who doesn't die just right, or yelling about something that doesn't explode in a way that is worthy of one of his performances.
Also, and I think I'll get skewered here, I think the laugh should be fairly low most of the time. Occasionally it goes to a cackle, but it should be rare so it really strikes through important moments. Kind of like a laugh that rolls along wondering whats so funny until the crazy takes over and makes every sick and twisted deed hilarious. If anything its more important to me that the laughs be abundant rather than high.
Also, I would like to see Joker generally presented in more full body shots. His clothes, his body, and movements are all significant parts of his character and I would hate to see those discarded in favor of twenty close-ups on the smile.
