1) A full, active, realistic, scale, destructible Manhattan and Queens, with many enterable buildings and at *least* 3 dozen distinct character models with many palette changes.
2) The best of all previous Spider-Man games, SM2's swinging with a hint of USM and USM's bouncy combat with a healthy dose of SM2's combos, Neversoft-calibre unlockables and replayable with one or two villain alternate storylines (ie Venom and Harry).
3) Peter Parker's life. I want a Camera function ala BG&E or Pokemon Snap where you can get money/unlockables/pictures/concept art/comic books/awards/whatever for taking pictures of the right things at the right times. I want to be able to talk to MJ, just like any RPG character talks to their female interest. I want some missions, preferably side missions to be based entirely on the relationships in Peter's life, MJ, Gwen and Aunt May especially. I want to have multiple clothing sets for old Pete and for MJ to play a notable part in the story and action, just like she does in all the movies. Spider-Man is about Peter Parker, the comics get it, the movies get it... it's time for games to catch up.
4) A taunt button. This is what would make Spider-Man SPIDER-MAN for me. I want a button to press that would unleash a quip at the target, the voice over would say something deroggatory at the opponent. There would be maybe 30 or so lines for the common criminal, perhaps lifted right from comics. There would be 5-10 for any given boss or special infantry type. And perhaps a dozen or so for special bosses.
Not only will it make Spider-Man fun and Funny BUT it will also trigger the enemy to attack quickly and wildly in anger. This can be used to protect innocents in the area by drawing fire, draw enemies in your direction for a trap or to get them to attack whatever's behind you. Some villains would be too smart, but ten there's be that back and forth that makes Spidey comics so awesome.
Some might say it could get a little repetitive... I say: hey you're doing the same ten attacks on every villain the game... what's wrong with having the same 70 lines that you only have to hear when you want to?
There's more I Could say, but having those four things would make the game the definitive Spider-Man game.
