Questions on Multiple Partitioning

I'm going to college for a degree in MIS, so I'm trying to learn how to use Linux, and might end up using it as my main OS after I get better at using it. My MIS teacher told our class about doing multiple partitions on a hard drive, and that its great for keeping data and increasing speed. I've partitioned my hard drive before when reinstalling Windows, but it was always on a single partition.

I'm just not sure how multiple partitioning works. I know you can separate your OS on one partition and have program files on another, but is there some applications that need to be in the OS section of the partition? Does anyone know a really good website to explain how to do this?
