Brief Introduction:

This is some older news pertaining to who this mystery character Karen could be whom I guessed before could be Margot or her mother (supplanting the father that appears in the comics perhaps):
Exclusive: 'The Eternals' Mystery Character 'Karen' is Revealed! - MCU Cosmic

In the comics, Margot Damien doesn't really fit the strong leader type the casting calls for. Her mother as far as I know is never referenced in the comics just the father but the movie of course will take it's liberties on this perhaps depicting the Damien family as more diverse and modernized. Therefore, the archeological expedition Margot's father is coordinating at the beginning of Eternals Vol.1 could in the movie be coordinated by her mother who knows...

Also in the comics, Margot of course becomes Ike Harris's girlfriend as well (Ikaris modern human identity of a photographer working on the archeological expedition alongside the Damien's).

Here's the comic history for reference sake:
Within Volume 1 there were a few loose tie-ins to the 616 universe through reference of both the Shield organization and the Hulk character that seem to have been forced by editorial before Volume 1 abruptly ended. Therefore, it seems odd that Dr. Daniel Damien actually appeared in Fantastic Four Vol.1 #64 in 1967 which is about 9 years before Eternals Vol.1 #1 even came out.

Daniel Damien's appearances in total are:
latest

*1967: FF #64
Doctor Daniel Damian was an archaeologist and father of Margo Damian. During an expedition on a tropical island, he discovered a hidden Kree base, guarded by Sentry 459. He and his assistant were imprisoned by the Sentry and saved by the Fantastic Four

*1976: Eternals Vol. #1-4
Later, Daniel hired a cameraman named Ike Harris (actually the Eternal known as Ikaris) to guide his daughter and he through the ruins of an Incan temple in the Andes Mountains and document the event. When they arrived at the ruins, they discovered an immense God Chamber filled with artifacts and technological wonders unlike anything else found on Earth. Ikaris revealed his true heritage to Doctor Damian and his daughter and activated a cosmic beacon to summon the great Celestials. Damian was present when the Deviant overlord Kro invaded the temple and challenged the approaching "Gods"[2]. He was also witness to the resurrection of Ajak and the arrival of the Fourth Celestial Host[3].
Arishem the Judge set down in the City of the Space Gods where it was decreed he would stand in judgment over Earth for a span of fifty years. Upon his arrival, the City of the Space Gods was sealed off from the rest of humanity. Ikaris and Margo Damian managed to escape before they were trapped for half a century, but Doctor Damian, unwilling to see the treasures of the Incan past lost forever, elected to stay behind

*1989: Thor Vol.#1 291 (1 issue of long ongoing Thor: Eternal Saga that first appearance of Eternals after Vol.1 ended)

*1991: Eternals: The Herod Factor
After the murder of his daughter Margo, Dr. Damian wanted revenge against Deviants and Eternals. To this end he planted false prophecies about the twin offspring of Eternals and Deviants conquering the 2 races and mutated Ajak into a monster that began killing human sets of twins. Later he had Thena and Kro's twins Donald and Deborah Ritter kidnapped in the hopes he could make their parents witness their demise. Ikaris discovered his friends betrayal and with the Eternals confronted Dr. Damian. Ajak disgusted with what Damian had turned him into killed himself and Damian by exploding.

She becomes Ikaris's girlfreind through the course of these issues.
Margo Damian was the daughter of famous archeologist Daniel Damian. She accompanied him and a cameraman named Ike Harris (who turned out to be the Eternal Ikaris) on an expedition where they discovered the existence of the Celestials, Deviants, and Eternals[1]. When the Celestials sealed off the City of the Space Gods, she escaped with Ikaris while her father stayed behind. Ikaris left her in the care of Sersi while he went to fend off the Deviants' attack on New York City[2]. They were both captured by Kro's forces[3]. When Thena and Kro agreed on a truce to reveal their races' existence to humanity, Margo suggested that they seek out Professor Samuel Holden, a colleague of her father

Yeah that’s a good question

I mean once upon a time I would have said Jonathan Rhys Davies for Daniel and Vanessa Kirby for Margot but I think marvel want to broaden out the casting a bit. I’ll have to have a think about it
