Xena and Gabrielle and Agron and Nasir probably top my list. As a g@y nerd, the depiction of Agron and Nasir on Spartacus was pretty refreshing (and revolutionary) for me. The fact that they received a "happily ever after" ending was also pretty monumental (especially compared to the ending of Xena: Warrior Princess).
