You'll notice after you draw that the aiming reticule cycles between red and white; try to time your RB/R1 presses when the reticule is white, this fills up the bar faster. Also aiming for vital spots like the head or their gun hand will fill it up much faster. It's possible to disarm (or maybe kill, tho I haven't pulled off a one-shot kill) someone with one shot if your timing is really spot on.
I normally wait until they've drawn before I do, if you time it well you'll still shoot first. I think I read that drawing after your opponent makes you more accurate but I can't find the article now. If you're having trouble with someone (Cornelius Wolfe killed me several times before I got the hang of it) try shooting their gun or their hat instead.
If you want a one-shot kill, I've found when it says draw that if you pull down on the right stick, then draw the pistol and target the head/arm for a kill/disarm and paint it exactly when the reticule is white you'll hit your target bang on.
