Nah. It's pretty much agreed that that's just a comic cover and someone slapped 2012 on it. It does say Comic News at the top of the page.

But it's on Marvel's official little hub for the movie, comic covers are never made in those dimensions or are just patterns slapped together.

It's from Mythos: Spider-Man, which was a painting retelling of Spidey's origin (art by Paolo Rivera). It was really nice, too.

In my opinion, that image is concept art for the opening credits of the reboot.
