Probably one of the most important themes and/or scenes of this movie is going to be the Weapon X adamantium-bonding operation where he gets his claws --- its the moment that permanently alters Wolverine into the man he is today, and the character we see at the beginning of the first X-Men movie.
The question then is what part of the movie should this operation be shown at, if it is shown (which is most likely)? It makes all the difference, because whichever act the operation occurs in will influence how the other acts of the movie play out.
I'm leaning towards having it in the middle of the movie; this way the first half could be all buildup, with the second half showing mass carnage as a wildly animalistic and vicious Wolverine rampages through the Weapon X facility in a way similar to how he did in the Barry Windsor-Smith Weapon X comics.
