Not a definite, but he will probably unlock pretty quickly. It really depends on how they set up the story levels, and how they set up the open world, to how important that is.

In Lego Batman 2, you didn't get Superman until about stage 6 or so i think, but you couldn't switch who you were controlling in the open world parts until you finished story mode. Marvel changed that in that you could switch characters at SHIELD hubs, but it still took time to get certain characters.

Lego Marvel unlocked characters in the following order, so you can probably anticipate something similar.

Stage 1 - Iron Man (mark 6), Hulk/ Bruce Banner/ Spider-Man
Stage 2 - Captain America, Mr. Fantastic
Stage 3 - Hawkeye, Black Widow
Stage 4 - Wolverine
Stage 5 - Iron Man (mark 1), Iron Man (mark 42)
Stage 6 - Human Torch
Stage 7 - Thor
Stage 8 - Cyclops, Jean Grey, Storm, Iceman, Beast
Stage 9 - Thing, Invisible Woman, Nick Fury
Stage 15 - Abomination, Green Goblin, Venom, Doc Ock, Mystique, Magneto, Doc Doom (you may unlock these guys in story mode for "purchase" earlier tho)

I feel like a little kid with how excited I am for this game.

I would think they would do a team-up level with the Teen Titans, like they did with the X-Men and Fantastic 4 in Lego Marvel. But what do I know. Cyborg and Robin both have what seem to be big roles in the game and Beast Boy has been shown as well.
