Offhand, I can think of:

1) Margin Call: 4 actors: Kevin Spacey (Lex Luthor), Paul Bettany (JARVIS, Vision), Stanley Tucci (Abraham Erskine), Jeremy Irons (Alfred).

2) The Horse Whisperer: 2 actors: Robert Redford (Alexander Pierce), ScarJo (Black Widow)

3) A Love Song for Bobby Long: 3 actors: John Travolta (Howard Saint - The Punisher), ScarJo (Black Widow), Gabriel Macht (The Spirit)

4) US Marshalls: 4 actors: Tommy Lee Jones (Harvey Dent), Wesley Snipes (Blade), Robert Downey Jr (Iron Man), Joe Pantoliano (Ben Urich - DD)
