616 is the cosmic designation fer th' main Marvel Universe, where most of th' comics and events take place. Just a way to classify realities.

It was almost done in Ultimate Fantastic Four...but it was later discovered the Universe Ultimate Reed Richards found was not 616 but rather another universe where everyone, including most of the heroes, were Zombies...

...to answer your question, I doubt it....but I've learn to put nothing past Marvel, so who knows?

Thus far no, it has been stated many times by Marvel that the Ultimate Universe and 616 will never meet. We're hoping that stays true.
