How should they portray Kryptonite? Random crystal, radioactive material, or drug that mutates humans?
 
I like a good ol' chunk of radioactive rock, sourced from inside the planet Krypton and fllung through space by its explosive demise.

I think what's more important though, is how it effects him. I don't mind Supes still putting up a but of a fight at first, but I do like my Green K painful & debilitating!

It should have the same effect as Uranium does to humans... Slowly liquifying your internal organs.

Visually, I want it to obnoxiously glow at all times, like the Tesseract in the MCU. It should look like something that doesn't look safe to touch even for a human. That sounds like a given as most live-action depictions have made it glow, but there have been a few that have went without.

As for it's effect, it should be crippling & physically hurt him, my favourite depictions are when it quite literally puts him down rendering him virtually helpless when he's near any sizable chunk of it.
 
Crystals, but rarer than yellow diamonds.
I'd make it that less than a half a.pound is on the entire Earth.
