I think it will be an alien lifeform, much like The Animated Series. Raimi derives a lot of the movies from that source as much as he does from the comics, so it wouldn't be a surprise. I doubt a moon expedition is how it will go down, but a meteor from space holding an alien lifeform doesn't sound like a bad origin to work with.

It seems likely, although for cinematic purposes I believe that it would hit during the night and not during the day.

I personally like how Raimi decided to use 2 villains who are able to alter their shape to some extent. I automatically assumed that their origin was somehow linked, something along the lines of sandman entering the testing "particle physics lab" where they were testing either the "prometheum X" or the symbiote itself. The symbiote escapes and Sandman is able to recombine himself in much the same matter that the symbiote is able to.

That was the intial idea that came to mind when I first heard about the 2 characters. As much as I like realism when it comes to super hero movies, (no other dimensions, galaxies, gods, etc..) I really would be somewhat upset if they didn't do the John Jameson thing.
