The Amazing Spider-Man Donald Glover wants to be Spider-Man

People can do fake accents. They can act American. How is Glover suppose to "act" white? Are they going to paint him white head to toe and give him a whig? That is equivalent to a non-American playing an American. The accent works, the painting does not.

They would have to do heavy makeup, which is possible, but would be time-consuming and hard.

Now, were they going with a Beowulf/3D mo-cap kind of thing, I would have no problem with Glover, because they can make him look the part.

He's too old - The studio is looking for a highschooler, Glover's 27, to quote Yoda, "He's too old"

He's not that good of an actor - He really isn't. Looking at imdb, he lost the role of Barack Obama to Fred Arminson, that's how much of a sucky actor he is, he lost a role of a black guy to a white actor.

He probably can't carry a big blockbuster - It's true, he's never had a huge blockbuster.

People won't accept it - again, this is fact

Things in the Spider-Man mythos would have to change - Yes they will, like Aunt May, Uncle Ben and others

Spider-Man is too great of an ICON to get a drastic change - This is not even debatable.

Why would anything in The Spider-Man Mythos have to change ? What their skin colour ? Why would that matter ? It wouldn't change the story or characters.
