I'd hate it if some guy was tossing out Joker cards before he fell into chemicals and became the Joker. The Red Hood origin is fine, but it cant happen in the film, unless it's through flashbacks. And even then, what's the point if Batman wasn't there, and it's kinda too late for that too.

But even then, i dont think the mask comment is literal in any sense. I dont think he's referring to face paint, or even prosthetics. Statements like "it's a beautiful mask to wear" and "any opportunity to don a mask is exciting" are about as metaphorical as they come.
