I think, hell, I KNOW Hamill's too old-looking for the part. I'd still like to see him play, simply from acting and personality alone, but as much as I clamor, the higher-ups undoubtedly would write him off as "too old." 
Glover's face with Hamill's voice would be perfect, but seriously, that's far more of a longshot than Hamill by himself (and just seems silly in general).
I don't know about Glover myself, but every movie that I've seen him in (he's had a few supporting roles as well), he's just got a whiny (not high pitched; whiny), unassuming voice and persona. Maybe he's got more range, but even in Willard, he was whiny.
I don't really know about Penn. He's an awesome actor, but he really does seem like a wrong fit for the Joker. His take on the Joker would be interesting and darker, but would it really be Joker? The character who even casual Batman fans have expectations for? Penn's a good actor, so if he somehow does happen to land the role, I wouldn't be too upset just as long as Penn delivers.
Realistically though, (that is, putting aside my obvious bias for Hamill), my favorites are Alan Cumming and Jude Law. They can look the part, they're good actors, they've shown dark and clownish (sometimes both) in previous roles, and they're big enough names to star opposite Bale.
