"Rune Thor" is one of the names given to Thor after he regained the Odinpower and learned to use rune magic in the "Ragnarok" story. Basically, he's the most powerful Thor ever. Here's the heirarchy:
Thor = normal Thor, one of the physically strongest characters, able to do lots of stuff with Mjolnir, able to control the weather
King Thor = Thor with the Odinpower but not a whole lot of knowledge on how to use it--basically he was normal Thor but stronger and with more energy/magic abilities
Rune Thor/Ragnarok Thor = the one I just mentioned above--full knowledge of the Odinpower, more wisdom than even Odin had, and mastery of rune magic in addition to his normal powers. He was like the Hulk, the Silver Surfer, and Dr. Strange rolled into one.

Yes, that was revealed. But then it was revealed later that there was indeed a real man named Don Blake whose life Odin basically hijacked and gave to Thor. Blake was stuck in suspended animation in a cave near the one Thor rediscovered Mjolnir in for years.
Which one's the real truth at this point? No idea. It's more than likely that, with Marvel's current editorial staff and tendency to ignore or forget continuity, the revelation that Don Blake was a real person will just be tossed to the wind and we'll find that this new Don Blake is somehow Thor again, even though Thor lost the Don Blake identity at the beginning of Simonson's run in the '80s.
