Considering that one of his crowning moments in the comics was a direct result of his constant chainsmoking... yes, yes it is a defining trait. As for the show. One thing really, really irks me, Constantine comes off as a bit of a suck up to heaven, whining that his soul is damned. Considering, that Constantine in the comics, despises heaven as much as hell because he considers them a bunch of litte tyrants and god a madman, him whining to the angel and searching redemption is a bit of a slap in the face. That and his trenchcoat is way to short. ^^
Being upset about the smoking, I understand. I personally don't give a ****, but I understand because that's his trademark and leads to a few important storylines (ones they probably wouldn't want to adapt anyway since they're doing their own thing). But the bisexual thing... Wasn't that mentioned like... Twice in the comics?

Sometimes, however, he just sees himself as one who must take the long, hard road to do what must be done.

Because of this, he is very rebellious, having a distaste for social institutions and norms and is disrespectful of rank and privilege, though he often has said rank and privilege himself. Wow that be cool just hope that his niece (Gemma) isn't introduced in season 1. I remember reading I forget which issue #275 or 6 I thinkwhere Demon Constantine rapes her and she goes crazy.
