I'm not saying I want to see any of them more than another, but there's:

Hydroman
Spider-Slayers
Electro


... and many more.

As a main, I think Mysterio could carry the main plot. Also, you could do many interesting things with Mysterio as far as effects go.

I think Electro could be used as a Sandman-esque side villain. (i.e. not as evil as he may appear to be)

Also, I'd never use Black Cat as a villain. A love interest, yes. Villain, no.

And finally, I'm debating on whether Kraven could be translated well on screen.

The Spider-Slayers could work, and throw in Alastor as the Ultimate Spider-Slayer in the end. Although, seriously, I wouldn't mind to see Mysterio. He could make Spidey re-face all his greatest foes at once in the forms of illusions! :P
