Spider-Man is my favorite. Also, you forgot Wonder Woman.
 
Ben Affleck's Batman, closely followed by Lex Luthor
 
Best - The Punisher, Deadpool and Spider-Man

Worst - Lex Luthor, The Joker, and Batman
 
1. Spider-Man (my favorite onscreen Spidey so far. He talks, moves, fights, looks exactly like Spider-Man should.)
2. Punisher (close second. Bernthal was awesome)
3. Batman (I guess, didn't really care for the other choices on the list. Alfred, Nightcrawler, and Colossus were good as well.)
 
Since TV is in play, I really like the new version of Silver Banshee. Granted, she wasn't in a previous film, but there was a version on Smallville. Siobhan's personality was developed quite well prior to her curse being discovered, and her grudge with Supergirl and team-up with Livewire (also a well-executed character) was an excellent payoff to that storyline.

Liked all of the ones I have seen, not seen GR yet. Loved the new iterations of X-Men and Spidey, but Bernthal as Punisher just kills it in every way. I have only seen 6 episodes of season 2 so far, and The Punisher is just amazing.
 
I enjoyed Joker's night club, tattoos, and him being kind of an antihero.

edit - While I liked Lex's backstory, I didn't like him being all manic - it seems like someone that went through his childhood would be more austere and distant, like in the cartoons.
 
Spider-Man with Punisher being a close second. I would give the edge to Spider-Man mainly because I've never liked any of the previous movie incarnations (and honestly downright hated the character in the "Amazing" movies") and enjoyed Holland's youthful energy in the role.

It's certainly not fair to favor a 15 min cameo over Bernthal's tremendous portrayal of The Punisher in 12 episodes but who cares. :D

PS: Needless to say that, aside from Iron's Alfred and Gadot's Wonder Woman all the DCEU "reinventions" ranged from "rubbed me the wrong way" to "made me feel nauseous" and I really didn't care for any of the "new" characters introduced in Apocalypse.

It's a tough choice between The Punisher and Spiderman. I went with Spidey because I knew right away I'd like Bernthal better as the Punisher, Spiderman I wasn't so sure about, but it ultimately won me over.
 
Wonder Woman is definitely missing from the list.

Going to have to say Ben Affleck as Batman, as the darkest and craziest Batman yet. The new Spider-Man was pretty great ( a very close second) as was Wonder Woman (third). Colossus was pretty good too, a perfect straight man for Deadpool. Alfred, I thought was well done (for a small amount of screen time, although to be honest he wasn't that different than Michael Caine's Alfred, just a bit more jaded).

I think I was one of the few who kind of liked Jared Leto's Joker - he took a lot of risks with the character, and Joker as gangster-pimp is a new thing. Sure, it wasn't even close to Ledger's performance (in terms of amazing acting) but at least it wasn't just aping previous interpretations. I hope that we find out that he has capped teeth because Batman has knocked all of them out. Suicide Squad was not a good movie, IMO, but I felt that the new Joker had some promise, which will hopefully manifest itself in the upcoming Batfleck film.

To be honest, most of the other characters on that list I found pretty forgettable (particularly the X-characters).

On a related note, if we were looking at TV shows, it would be Tyler Hoechlin as Superman by a mile.
 
Wonder Woman is only missing if you count her TV version.

Per the title "new take", meaning a character already portrayed more than once, I'll take Punisher and Colossus.
 
Spider-man but not a knock against Punisher at all. Jon was amazing
