I think it's not so much that the sense of humour is different (though that's certainly part of it) it's that you simply can't translate something like wordplay. You can explain why it's funny in the original but most of the humour is inevitably lost. And since the lip movements need to be in sync the characters often end up saying something that's little more than a rough equivalent of what was actually said.
It can really change the tone of a scene or turn a great joke into something so tacky that you feel embarrassed on behalf of the character who spouted the nonesense.

and that is why physical humor like the Stooges tends to "work" as it isn't based on that wordplay, true.
