Use one of the three and you have solid a great antagonist for the first film. Mysterio and Scorpion haven't been used yet and are pretty big villains. Especially Mysterio. Venom's story was never done properly and going with the Symbiote for the first film would be pretty bold.
I honestly agree, though I think the way to work Venom is to kinda use the cosmic Marvel movies to bring in the symbiote. That way, you wouldn't waste time establishing Spidey's world in his own solo film. You don't have to worry about coming up with a good reason why the symbiote exists. Just have him catch it in another film. That way, you can establish the symbiote, then, in his own film, establish why Spidey doesn't want it.
But Scorpion and Mysterio are two bad guys I'm really gunning for. Scorpion especially, if created as a weapon to destroy Spider-Man, can have a real existential crisis, or go out for revenge against Jameson--assuming he creates him--or even Smythe if they have Smythe invent his tech. Even better, if Norman Osborn recruits Scorpion later as a personal body guard or hired gun, kinda like the Thunderbolts, you can see a lot of potential there.
And Mysterio is Mysterio. He's just awesome.

Build up to GG. He's coming. There's no doubt they'll use Norman. But Doc Ock would be cool, especially if he's building up his own personal army. It's been long enough where we're due for another Doc Ock.
Still, I'd prefer if they built up to Doc Ock or GG, and not just introduce them immediately. Like, mention Octavius or show Oscorp and Norman. This is one thing I feel TASM got right...but kinda bungled along the way.

Oh, I think he should do a ton of damage, and maybe appear sinister at first, but, as the film goes on, and Spidey just relentlessly teases this guy, he is played more for laughs at times, but not in a way where you don't take the guy seriously. Rather, I'd like it to be that you kinda want to see this guy win a little bit just because he's having such a hard time. I feel that Shocker should be kind of a sympathetic villain--not in that you hear is tragic backstory or anything, but rather that he's trying really hard to get some money, coming up with all these cool gadgets and plans, only to be taken down by some teenager in a costume.
I guess a good comparison would be those execs from Cabin in the Woods, where you know they're doing some pretty horrible things, but they're so entertaining along the way you kinda root for them, kinda? I don't know if I explained my point well enough...
