I would have the movie begin with the typical SPIDER-MAN style credits. The credits flip and zoom around a large sphere with webs flying to and fro. Then as the camera moves back you see that the spere is really more like a giant egg. and SPIDER-MAN 4 comes on the screen just before the egg breaks up and hatches, and The Lizard flies onto the screen and sticks.

Adrian Toomes would be a business man who hired a master assassin, the Chameleon, to take out big business rivals. Since Spider-Man saved Gwen from being killed by that crane in Spider-Man 3, Captain Stacy has started working in conjunction with the web-slinger to apprehend these gangs being supplied by Toomes. Toomes is prepared for Spider-Man. The businessman has designed a flight suit with a number of features, such as wing-mounted homing missiles (the Feathers of the wings are the missiles/pin missiles). Spider-Man manages to apprehend Toomes and swings away before the NYPD can take him in. At this point they dont know Spider-Man is working with Captain Stacy. Its a secret. Confessions from various thugs, inventories of missing gear from Toomess company, and leaks about illicit human experimentation ends up putting Toomes behind bars.
Mary Jane has been having nightmares of Peter losing control of himself, combined with flashes of that time at the jazz club where he hit her. She decides to go to her mother for advice. She tells Mary Jane about how she had fallen in love with her father much like Mary Jane did with Peter. After this, Mary Jane begins to think about whether or not she wants to stay with Peter.
Peters grades have seen such an improvement that Dr. Connors has asked him, Gwen, and a select few other students to assist him in a research project. They are studying the regenerative effects of reptiles in relation to human physiology and biology. That night at the Connors household, the doctors wife asks him how much longer hell be doing late work in the lab. She reminds him about how hes missed important events for their son by working late in the lab. He tries to apologize to her, but she tells him to apologize to his son. So Connors goes to his sons bedroom and apologizes. Though the boy doesnt really accept the apology, but gives Connors more a thats okay and crawls into bed to sleep. In the hallway Mrs. Connors tells Curt that their son needs a father wholl be there for him. Then she tells him flat out that shell leave him if theres any more late night research.
The next day Peter goes to kiss Mary Jane goodbye and she accepts the kiss but flinches at the touch of his hand to her shoulder. Peter pretends he didnt notice and heads on to school. Mary Jane gets up from the table and digs out that play Harry had written and begins reading through it. At college, Connors has cancelled the days lectures to finish up his research in one long session. Many tests on guinea pigs yield fascinating results, such as hairless, slightly scaly re-grown limbs. It becomes evident that more research will be needed to finalize the product. Regardless, Connors actually heads home early, slightly disappointed that his research is still unfinished but the reception he receives from his wife eases the disappointment. Shes happy, smiling, and just glad that he made it home early. Just in time for their sons elementary team basketball game.
Peter returns to his and Mary Janes apartment to find her reciting lines in front of a mirror. She tells him that a production company bought Harrys play and offered her a part. Peter tells her thats great when there is a crackle of static from the police scanner. Peter suits up and swings out into the New York night. Following the call he finds that there was a break out at the prison. A crazy man with shockwave blasters uses them against the cops and busts down the northwest wall of the prison. Spider-Man and Stacy manage to apprehend him before setting out after the thugs set loose by the Shocker.
Peter returns early the next morning tired from the long night to find a note from Mary Jane stuck to the fridge saying she got a part in the play. After breakfast and several cups of coffee, Peter leaves for classes. More tests are run and what Connors believes to be a finalized product is developed. He congratulates the students who aided and dismisses them for the day. Gwen asks Peter what hes doing later; he tells her hes going to the new production at the theatre.
After the play, they go for a walk in the park where Mary Jane tells Peter about the play hitting the road, and how they want her to go with them on the country-wide production. Peter nods and tells her hes notice shes been acting differently lately. He asks if it had anything to do with that thing (meaning the symbiote). She reluctantly tells him that yes, its had her worrying and wondering what would happen if something else happened to Peter would he accidently use his powers in the wrong way would he accidentally hurt someone he loves Peter nods and tells her hes had similar thoughts. He then tells her that he loves her, and if she would feel safer away from him, then he wouldnt hold it against her. They share a hug under the moonlight and she thanks him. He wishes her luck in life. They kiss.
Suddenly something bursts from the foliage nearby. Peter pushes Mary Jane out of its way and tells her to run. He rips off his clothes, revealing his Spider-Suit beneath it. He pulls on the mask and leaps into a tree to search for whatever had followed them. His Spider-Sense goes off and he sees a rustling in the bushes nearby, following the progress of another couple. Peter nods at the rustling and tells Mary Jane she might want to get out there. He pulls off his shirt and pants to reveal his Spider-Suit, dons the mask, and stealthily approaches the thing following the couple. His Spider-Sense alerts him that its preparing to pounce. At the exact moment that the watcher makes its move, Spider-Man makes his. He collides with a large, reptilian monster. The Lizard. The couple runs off screaming as Spider-Man tussles with the Lizard. The fight ends with Spider-Man getting hurled into a tree and the Lizard fleeing. Spider-Man pursues the beast to a manhole and decides against tailing it. Instead he swings by the NYPD and waits.
When Captain Stacy leaves for the night, Spider-Man confronts him and asks him about any reports of animal attacks. Stacy shakes his head no. Theyre still searching for the Shocker. Spider-Man swings back to the apartment to find Mary Janes bags packed and her asleep.
The next day, Connors isnt in class. Gwen mentions that her father got some interesting reports early that morning about partially devoured sewage plant workers and a couple of joggers. Peter stops by the Connors house hold. Mrs. Connors asks Peter if hes heard anything, anything at all about Curt. He tells her no. He was wondering why the doctor didnt show up for the days lecture. The Connors child, Billy, tells Peter that he saw daddy outside that past night. Peter asks about that. Mrs. Connors tells him that Billy woke her up saying daddy was outside. She checked and saw no one. Figure Billy was dreaming.
That night Mary Jane leaves. After Peter sees her off, he swings by the Connors house and perches in a tree in the backyard. Soon the Lizard ambles into the yard. Spider-Man watches in silence as the Lizard sits before Billys window, watching his son with what appears to be some measure of intelligence. Afterward the Lizard leaves and Spider-Man follows him into the sewers. Suddenly the darkness is flooded with lights: the police lead by Captain Stacy has set up a trap to catch the Lizard. Lizard panics and begins tearing into the ranks of officers. Spider-Man must protect the Lizard from the officers, and the officer from the Lizard. Lizard manages to escape; Spider-Man follows him. Eventually they lose the police. Spider-Man follows the beast to a lair of sorts. The Lizard then turns and takes another way out of the sewers, leading back to the Connors house. There he smashes into the house and abducts Billy. The NYPD arrive, but by that time Billy is gone and Martha Connors is seriously wounded. Spider-Man returns to his apartment.
Peter makes plans to collect a sample of Connorss skin and blood. He then suits up and heads back into the sewers, where he finds Billy left alone and sleeping. He hears a splashing down the tunnels and hides. Lizard returns with a dead fish and lays it beside Billy. Then Spider-Man makes his move. He gets his sample and flees. Later that day in the lab he manages to concoct an antidote to the serum Connors took. He returns to the sewers and fights Lizard a final time, administering the serum. The Lizard becomes Connors who doesnt seem to remember a thing. He tells Connors about the transformation, about attacking his wife, about abducting his son.

No, lets just have it not be a bunch of accidental bitings this tim.e How about the Lizard isn't Curt Connors (since, as you explained it no longer can be) and have the new Lizard just come from an egg.I dunno why Raimi turned Curt into a particle physicist or whatever instead of a biologist but that pretty much alters the Lizard origin. So here's my take on a new one.
Connors tells Pete his symbiote sample starved to death but he in fact bonded it permanently to one of the lizards in his home lab. A sPecies skink of skink which reproduces asexually. He notes it's amazing regenerative abilities. The skink lays a bizarre egg.
Curt comes home one night to find his house has been broken into. Cletus Cassidy, a crackhead desPerate for money loots expensive looking equipment in his lab. A fight breaks out between the two and in the struggle, they crash into symbiote lizard's tank. The lizard, crushed under Curt's body, bites him as it's killed. Cletus smashes the eggs under his arm, and the mysterious embryonic symbiote burns itself into his skin.
Cletus escapes and the two begin their resPective mutations.

well, he did it in the SSM cartoon. I don't think it is the same thing. maybe he doesn't even feel his prosthesis. and have a arm prosthesis when everyone else around you have a normal arm may not be a cool experience.Oh, and I think there's a question that needs to be answered if you use Lizard: If Connors is doing experiments only in hope to grow his arm back, why would he take such risks and not simply use an arm prosthesis?
I'd like to see a movie with Lizard where Curt Connors is going through a slow transformation similar to David Cronenberg's The Fly. The monster in that movie was scary, but still you felt sympathy with it at the end.
Oh, and I think there's a question that needs to be answered if you use Lizard: If Connors is doing experiments only in hope to grow his arm back, why would he take such risks and not simply use an arm prosthesis?
