I have a story with my own team worked out as well. I'd start my fist movie with Mephisto using the Darkhold to create a new Hell on Earth. It's a lot more and I've thought out a trilogy of films, but I won't get into it here. First movie's lineup would be Strange, Blade, Ghost-Rider, Jennifer Kale, Man-Thing, Wolf by Night and Morbius.
Stephen Strange would lead. His well-rounded knowledge of sorcery, the supernatural world and not being an emotional wreck(at the moment) like most of the Sons makes him the prime case to lead, along with the fact that the Midnight Sons are using his Sanctum Sanctorum as a base of operations. Also the team medic, with his extensive surgical expertise. Strange has a strong need to save and preserve lives, by any means necessary. His methods don't always gel with the rest of the Midnight Sons, who don't all trust Strange as much as he'd like them do. Begins the film by attempting to solve a string of murders that are seemingly tied to Vampires.
Blade would co-lead as the team tactician. Has lead the Night-Stalkers before. Was on his own slaying Vampires in an attempt to solve an ongoing mystery, until Strange came to him, trying to solve the same case. Blade's been a killing machine for the past few months, so he's in a dark and not-so-trusting place when Strange finds him. Blade was near-death when Strange saved him, and had been recovering in Strange's sanctum for a time, while the world's remaining vampires believed him to be dead. Blade feels indebted to Strange, and the two share a friendship hinging on trust and necessity. The two agree to work together to solve a mutual murder mystery involving vampires. Still secretly dealing with his vampire-side, he needs improved serum to keep his urges controlled before its too late. Usually in control of every aspect in his life, Blade's not yet too trusting of this rag-tag team of monsters he now finds himself on. His no-nonsense personality, coupled with his unyielding work-ethic, works well to keep the team of monsters in focus of their mission. Blade's blunt words sometimes put him in closely confrontational situations with the more rebellious Midnight Sons, like John Blaze and Morbius.
John Blaze as Ghost Rider for his connections to Mephisto and his cousin, Jennifer Kale. Was unwittingly serving Mephisto under a powerful spell, until Strange broke him free. An emotional mess, constantly reeking of cigarette smoke and booze. John has aged well past his prime, constantly traveling to escape his tortuous demons and to maintain a safe distance from the few loved ones he still has. Always exhausted from a lack of sleep and faith, John is ill-tempered and rebellious; he wouldn't even be with the Midnight Sons if Strange hadn't saved him from Mephisto's spell, and more importantly, the life of his cousin. A soul battered by loss and grief, John might have a chance of piecing his life back together when he reconnects with Jennifer. No longer lets anyone but Jennifer call him "Johnny" anymore, since his younger days are well behind him. As a lone wolf and an atomic bomb just waiting to explode, Blaze bears a long-standing hatred for Mephisto and believes he should be the only one tasked with taking the demon down. Strange warns Blaze that going after Mephisto on his own could result in him being put under another spell, so Blaze remains with the team as a means to an end.
Jennifer Kale for her connections to the Darkhold, Blaze and Man-Thing(I'd replace her Tome of Zhareed-Na with the Darkhold). A young woman saved from an oppressive foster system by her older cousin, Johnny: Jennifer had a great life after that point, until she secretly witnessed her ailing cousin make a deal with Mephisto in exchange for his soul, later riding off into the night leaving flaming tread-marks. Johnny never returned. Kale wondered what happened to him until the mysterious Darkhold showed up on her doorstep, answering all her questions about what Blaze had become, as well as binding itself to Kale's soul. Was on a journey to find her missing cousin until she was captured by Mephisto's forces and brought to Mys-Tech, in order to use the Darkhold for their own agenda.
Ted Sallis, the Man-Thing would be there, a young college professor who fell in love with Kale, only to get murdered in a car "accident" and left for dead in a swamp. Jennifer's raw emotions over his death intermix with her connection to the Darkhold, and she subconsciously brings Ted back from the dead as the mysterious and lumbering Man...Thing? He shares a deep emotional, mental and spiritual connection with Kale, and will do anything to protect her. Kale still sees him as Ted, so they share a bizarre romance of sorts.
Jack Russell, Wolf by Night. Jack Russell was attacked by a werewolf one night. Although hideously scarred, miraculously, Jack didn't die; but he was transformed into a monster. Jack's subconscious mind took him to his home, where he murdered his family in a wild rage. After realizing what he'd done, Jack left his old life in the UK behind and moved to America, working odd jobs until he began brawling for cash in underground cage-fighting. Self-destructive and suicidal, Jack believed he deserved all of the pain and carnage that he was constantly dishing out as the Wolf. It's not until he was captured by Mys-Tech that Jack's wolf side was manipulated and forced to do Mephisto's bidding. Jack, along with Jennifer Kale, are rescued by the Midnight Sons when they infiltrate Mys-Tech. Jack feels like he has a one-way ticket to hell after what he's done as the Wolf, but stopping Mephisto might be the salvation he needs to guarantee his place in Heaven alongside his dead family. An Irishman with a strong pride in his roots, Jack loves drowning his troubles in booze with John Blaze.
Michael Morbius. Afflicted with a deadly disease, Morbius underwent a blood-transfusion procedure that turned him into a creature with Vampire-tendencies and abilities. Feeling like his wife and children wouldn't accept him after witnessing the creature he had become, Morbius felt forced to hide away from them and left his old life behind. Originally working for Mys-Tech as a human, Morbius began his new life as a vampire by finding acceptance through Deacon Frost, who welcomed him with open arms, despite Morbius not being a "true" vampire. Morbius ran with Frost for years, even fighting Blade on occasion, until he had a change of heart that forced him to flee Frost's ranks and retreat into the shadows. Morbius no longer wanted to feed on innocents, and instead began to survive on the blood of rodents he'd scavenge. It's not until Morbius' former peer in the medical field, Stephen Strange, comes asking to help that Morbius leaves the rodents behind and hesitantly joins the Midnight Sons, despite a clear rivalry with Blade. Morbius' knowledge of Mys-Tech, coupled with his past friendship with Deacon Frost, make him a team asset. He will later be responsible for creating Blade's new and improved serum, after the two come to a mutual understanding for each other.

Mephistopheles is the offspring of Lucifer and Lillith, and was effectively the first demon in existence. After a failed coup against Lucifer to take control of Hell, the Arch-demon Mephisto was sent "upwards", and banished to Earth. He was then locked in human form, cut off from most of his inherent magic and tasked with collecting souls for Lucifer. Once Mephisto had collected enough souls, he would be allowed back into Hell where he'd regain his lost powers, along with his Hell-Lord status and the respect he'd long since lost amongst his demon brethren. After being on earth for so long and converting obscene amounts of souls for his father throughout the ages, Mephisto now tires of his task, fearing that his father lied to him. He has massive power on earth through his manipulation of humanity and world events, but he craves his "real" power. He's now taken it upon himself to create his own Hell on earth, where the dimensional barriers separating our world and hell will be shattered completely. Once Mephisto comes into contact with the energies of his home dimension, he will regain his magical abilities and bring all of hell's demons onto Earth, stealing his father's subjects and creating his own kingdom for demonkind. Runs his company, Mys-tech, from the shadows. They specialize in secretly melding technology with sorcery and the collection of magical artifacts. Although the company is highly successful and their products are used worldwide, most people have no idea that Mys-tech products are made with magic. Mys-Tech is publicly run by a boardroom of secret immortal sorcerers, their souls owned by Mephisto through contractual obligation. The sorcerers also closely guard the depowered Mephisto to protect him from threats, while carrying out his plans using magic that he is no longer able to wield.
Deacon Frost, along with the rest of the earth's ruling Vampire elite, would also feature in the film. Being forced to work and murder for Mephisto under threat of being destroyed, the vampires are looked down upon by demonkind and treated as impure "sub-demons". Mephisto can't manipulate the vampires with his usual methods because they have no souls, so he threatens the annihilation of their entire race with the Montesi Formula, a spell found in the Darkhold. Wanting to preserve the Vampire race by any means, Frost and the Vampire elders have no choice but to begrudgingly serve Mephisto. Frost shares a deep friendship with Morbius and bears a long-standing rivalry with Blade. Has a weird "fatherly" relationship with Blade because he feels responsible for the Daywalker's creation. Blade despises his connection with Frost, and even hates it when Deacon constantly calls him "Eric" to remind Blade of his rough childhood.
