I'd establish the FF themselves via guest appearances/post credit scenes/etc in other films, then do a solo Doom movie. That's where we'd first see the origin in full only with Victor as the protagonist. (I'd keep as much to the classic 'Doom blames Richards for an accident in college' as possible). The rest of the film would deal with Victor-as-badass-antihero traveling the world and eventually conquering Latveria and becoming Dr. Doom.
THEN I do Fantastic Four, and even then I might use Mole Man instead just to piss everybody off(but serious: giant monsters!). But my goal would be to set Doom up as someone the audience kinda roots for, because he's awesome and super classy, but also feels guilty about it because at the end of the day he's a full tilt badguy, and as he comes into conflict with the FF (and later the Avengers!) that point will be hammered home.

So, it seems to me that there is ample opportunity to include the Fantastic Four in the MCU...assuming Marvel acquires the rights within the next year or so.
Given that the two Infinity War films will likely be absolutely massive, there's plenty of room there to introduce the FF and touch on their origin. Hell, they could even tie the origin into the events of the films themselves.
Following Infinity War: Part II, you've got the Inhumans, a property with very close ties to Marvel's first family. They could easily be included in some capacity in that film, especially given that the Inhumans were introduced in the Fantastic Four comics. Plus, it would provide an effortless tie back to Earth.
At this point, the Four would be well established over several films in the MCU. Assuming the public has now warmed up to Marvel's rendition of the characters (and there's no reason to believe they wouldn't), Marvel would likely feel comfortable featuring the team in their own film in Phase 4. This would be the perfect place to introduce Doom!
Plus, just imagine the villains that Marvel would be able to use to cap off their next several phases!
End of Phase 4: Skrull Invasion (with Super Skrull!) or even just Doom
End of Phase 5: Kang the Conqueror
End of Phase 6: Galactus
The possibilities are endless.
Here's hoping Fox will do what's in everyone's best interest and make a deal!

There is not the slightest chance in hell of Doom appearing on Netflix. It'd be just as expensive as doing an Iron Man netflix show.

Why? Doom's powers and magic stuff don't particularly demand any heavy CGI. Espically for an origin story in which he won't even have full powers until the final episodes.
Anyway, here goes my own personal plan for an MCU Fantastic Four.
Doom: A Netflix Original Series: Set in the late 60's, we see the origin of the Fantastic Four through the eyes of Victor Von Doom, heir to the throne of Latveria as he travels on his own journey. Besides Reed Richards and co., there's oppurtunities for cameos by the likes of Howard Stark, Peggy Carter, etc..., and roles for lesser known magical characters like Dr. Druid. Doom could even meet the Ancient One, if Tilda Swinton was willing to reprise the role. Series ends with Doom imprisoned and the F4 vanished.

Andrew Scott: Victor von Doom - Heir to the throne of Latveria, Victor made history by choosing to study abroad in a seeming gesture of good will between his homeland and America. A master of astronomy and quantum physics, he collaberated with Dr. Richards only to be forced out of SHIELD's programs by Dr. Franklin Storm,, due to his increasing interest in "unscientific arts" and infatuation with Dr. Storm's daughter. He disapeered, and shortly thereafter, a CIA-backed coup killed his father and took command of Latveria. And enraged Victor resurfaced on the day of the mission launch, assaulting SHIELD with new found powers before being captured. It is believed his attack caused the damage that caused the mission failure. The disaster was covered up as part of the space race, and the nation mourned the loss of four heroes. Dr. Storm resigned from SHIELD and disapeered. Victor von Doom has remained imprisoned in a SHIELD dark site ever since.
For 40 years, the truth about Dr. Richards' final mission has been kept a secret from the world. But in 2020, the Four are coming home.....
