It won't take up the entire film. There was a report mentioning that Joker will recieve a backstory. And that we shouldn't expect to actually see him, until the middle of the film. Like how Batman was shown in BB.

ok so "Philip Hoffman Seymour will Play Penguin and Ryan Phillippe is in talks to play Harvey Dent" is old news?

Even though Hoffman has been rumored for Penguin before, recently, his name emerged again, with the confirmation of Penguin being the "second" villain of the sequel. Though it's still being looked into whether both rumors are false or not. Phillippe is the current actor being attached to the Dent role. Which was reported over a week ago.

Goyer mentioned before that the sequel would be about Batman, Gordon, and Harvey, bringing in the Joker. Then toward the end, Joker would be the one to throw the acid. Setting up Two-Face for the third. Though it's possible that it's still likely to happen, things could easily change. Considering this was before Nolan's brother was confirmed to pen the script.
