Here are some more details from Kotaku:

The official Assassin's Creed web site gives us a brief taste of the second entry in the series and clues us in on when we can expect concrete details on Assassin's Creed 2—next Thursday.

The teaser consists of little more than a short Flash animation, rendered in a hand drawn style noticeably similar to that of Leonardo da Vinci's inventions—focusing on the concealed blade Altair used in the original—and his Vitruvian Man. The final drawing hints at some sort of winged contraption that may indicate we'll be doing some flying (or at least some floating/humming of the Batman theme song) in the sequel.

The teaser also notes that we'll see more in the upcoming issue of Game Informer, with the date April 16, 2009 presumably indicating when the games mag is allowed to lift the veil on Ubisoft's sequel.

Given the Italian pedigree of the teaser, we'd think those rumored details of Assassin's Creed 2 taking place in Venice are sounding more and more likely. We're sure to know by next Thursday. Keep an eye peeled!

I loved Assassin's Creed and am excited for this. Hopefully they'll add more variety, but it didn't bother the first time. I hope the new assassin has as cool of a design as Altair did.

Im with you. Id love to see Altair return but it doesnt look like thatll be the case. Maybe this new assassin will be tied to Desmond/Altair in some way.

I think it will be another ancestor of Desmond. Venice seems like a great location to do some free running and some kind of gliding device sounds awesome.

I hope they do better than what they had to offer with AC1. I learned my lesson and most likely wont touch this new one until its in the bargain bin

All they have to do is add more variety to your mission structure. I didn't find the game repetitive until my 2nd playthrough, then it hit me like a brick wall. Give us more ways to eliminate our targets, more ways to approach our target and I'm sold. Take some cues from the Hitman series and they will have another winner.

Yea i cant wait to see Venice. The Holy Land was absolutely gorgeous so i can only imagine how this will look.

The president of Ubisoft stated "before March 2010".

I'm guessing it'll be a November release. Id assume it'd be out by the holiday, just like the previous one was.

I hope we get to battle another assassin. One who is equally skilled or more so with a sword than you are.

But like i said, i just cant wait to see the world we will be inhabiting, the Holy Land was what drew me into the first game and it was just amazing.

Yea that does look alot like Altair. If anything it looks like the character is part of the same clan that Altair was part of.

Damn, im bout to go do all of that myself, thats pretty cool. So its deff part of the same guild/clan whatever you want to call it, but yea Altair was deff missing a finger, that guy not so much. Im gonna say this is another of Desmonds ancestors, i know they are gonna give us more info on Desmond so im sticking with that theory.

Ill say it IS desmond. the assassins where comming for him at the end of the first one so i think the rescued him. or maybe they will jump from past to present(future) but instead of walking around in a lab you have assassinations as desmond.
