No. No. No. No. No. Spidey should not quip at all when facing a major villain. Maybe once or twice during the very first encounter between the two but just let it go after that point. It would detract from the gravity of the situation. If there's a hulking, 9 ft, lizard beast threatening the lives of hundreds of my peers, I don't think I'd be making a whole lot of wise cracks. The way I could see it working is like this:
Spidey and Lizzie meet for the first time and Spidey makes a few jokes about how someone possibly turns into a lizard etc. etc. then mid-joke the Lizard roughs Spidey up and Spidey realizes that this isn't a petty thief like the ones he's used to dealing with. We maybe get a close up shot of Spidey whispering to himself about how serious this really is and whatnot and then that would be it for joking around when it comes to dealing with the Lizard. The jokes and wisecracks worked in Spectacular Spider-Man because it was a tv show with a target audience of younger kids. This film is going to be PG-13 with a target audience of 15-21 year olds. I don't need jokes every 4 seconds.
That being said, the complete lack of Spidey's humorous nature in Raimi's films was a bit odd. I mean even during the montages of when he's taking down thieves it was lacking and there was a HUGE opportunity for Spidey to dish out the quips when he saved Mary Jane which was totally wasted.
