I doubt that. The Lizard and Kraven make a great pairing. The Lizard and Vulture just sounds too weird.Yeah... we will probably get some oddball pairing like Lizard and Vulture.
1. Would depicting Dr. Connors as a main character be a problem because of how Otto Octavius was changed to be more "Connors-like" in SM2? There'll be similarities, but here Connors wife won't die or his son as well. Raimi could maybe change it where in The Lizard's blind rage, not Connors, he accidentally kills his own family, and than like a werewolf he'd change back in the morning, returning to his house, only to find his wife and son brutally murdered. The police would believe some kind of animal came in and got them, which thus would strike Connors into trying to find a cure and redeem himself for his family's death.
2. What's up with that "I'm a physicist, not a biologist." line from Connors in SM3? The symbiote sparked the idea of him creating a regeneration serum that could regrow his arm.
3. Since a Lizard vs Kraven vs Spidey storyline is used in the Sony SM3 video game, does that mean that there's now less chance of it being used in an actual movie? The games would not have an affect on the movie. After all, Vulture was in the first movie game, but Raimi wanted to use him for movie 3. Now, of course Lizard is in the 3rd movie game, but everybody seems in agreement that he is movie 4's villain.
4. How should the Lizard's transformation and possible cure be handled in a film version? I agree with someone who said that the cure should only be temporary. The cure genetically could relapse and return back to his normal human self, but like "X3"'s ending with Magneto, Connors could like look down at his good arm and start to see little scales forming and sprouting out of his skin. I also think his transformation should be like what we saw in MTV's Spider-Man series where he makes a video-logue before testing out the serum, he then tests it, we see lightning flash and silouhettes of him changing, before finally we see a desk chair thrown at the camera, test tubes and beakers thrown off the desk and smashed on the floor, and than a scaly-tail leaving the lab room. Throughout the movie we'd see little glimpses of his snout, eyes, fangs, and claws, before finally getting a encounter with him in the sewers.
5. What secondary villain, if any, should be featured with the Lizard?
Kraven, followed by Scorpion. I really want to see JJ totally fed up with Spidey and pulling all the stops and money into bringing down the wallcrawler. Maybe he could think Spider-Man is responsible for The Lizard and thus sends Kraven and Scorpion after the both of them.
6. And of course, what should a movie Lizard look like? Tall, should have the ability of other lizards. I'd love to see where the Lizard could adapt to surroundings like a chameleon and hide, I'd loved for him to have the speed of a frilled lizard, I'd love for him to have a poisonous spit and tongue, and I want him to look like a cross between a raptor and a crocodile. if they can do all of that than mission accomplished.
Dr. Connors Character has already been established for the past 2 Films. Spidey4 is the right time for Dr. Connors to become the Lizard. Sam even said Himself that with Dylan Bakers patience He deserves a Villanous pay off. And quite frankly I couldn't agree more with Sam. You can also count Spidey1 for the establisment of Dr. Connors because afterall they did mention Him in Spidey1. Dr. Connors even had Lizard experiments in His lab which hinted the Lizard. There really is no need for more establishment so us fans think it's time for him to become the Lizard. I think Eddie Brock was established perfectley in Spidey3. they told His story and then in the final battle they gave us Venom which I thought was awesome. For the sake of the film and characterisation, I feel Conners should be more established first, like they should have done with eddie brock. So have Conners as a more important character in SM4, and slowly show him dabbling into regeneration etc(even though he's a physicist in movie-verse????), and then the impact of Parker's friend and mentor being reduced to a beast would be a lot more worthwhile, and have that happen in SM5!
Have two villains in SM4, maybe even a third who is a throwaway token, as in an action sequence at the begining etc. Then, have just the lizard in SM5 and show the emotional and physical implications, we should see him family and things in SM4, so we get a feel he's a character, not a plot device.

I don't think fighting a "guy that helps you out" is worse than fighting your best friend.
The Lizard is (most likely) not going to be the under-developed character if there is one in S-M4. There's not even that much to develop left. We already know they have a relatively close relationship for teacher-student. Put the family in one of the first few scenes. Work with PP on the serum. More family? Use the serum. Change to the Lizard. Smack people up. Remorse for actions while being the Lizard slash not wanting to harm family switching back and forth with bloodthirst. Actual bloodthirst. Spidey looks out for family, etc,.
With a talented screenwriter and director, its pretty cut and dry. If need be, stick Kraven (or another baddie that can be connected to the story or theme) in there. But IF there's another villain, they will probably be the underdeveloped one.
Regarding his transformation, I'd like to see him grow more monstrous throughout the film. Starts out looking a bit like a sleestak, progresses to the Ditko design, moves onto the Ultimate look before culminating into something resembling the MTV series version. The idea of a 20 foot long reptile chasing sPidey across the city skyline kind of appeals to me. Plus, I'd like to see an action sequence at the Zoo.
And yeah, Kraven would be my second choice.

The Lizard is not teaming with anyone. He is out of control rampaging in the city. The dynamics and uniqueness of the story is its a villain against a villain against Spiderman. It is very different and something we have not seen. Kraven trying to kill Lizard and Spiderman trying to help him. Spiderman is caught in the middle. And Chameleon and Kraven should never have a relationship[ at all.I can't see Lizard pairing up with anybody to fight Spidey. And with all the talk of Kraven, why wouldn't we have him and Chameleon fight against Spidey together? I mean, they ARE half brothers, so Raimi could do his own little family segment on them and then possibly form some sort of plot around that...
Who knows, just throwing that out there...
