In terms of the mythology, Clarice is important. Her presence on the show would arguably be a bigger reboot than this season has been, because we'd essentially be getting a third protagonist. You can make alterations to the source material, ( for example, Fuller has suggested possibly changing Clarice's race) but I think it has to ultimately keep true to the spirit of the novel. Mason Verger and Cordell didn't try and personally cannibalize Hannibal in the books or the films, but it still feels true to that storyline. If they did Silence of the Lambs they'd need to keep that Hannibal/Clarice element intact because ultimately that's what the story is about.
But I think they could push on without that storyline if they really wanted to. Fuller has said time and again that he wants Lady Murasaki introduced into season 4, so maybe if he wants to touch on Hannibal finding companionship/friendship/love outside Will he'll explore that character.
