I think the MCU Defenders will basically take up the role of the Avengers in the early comics. A more street oriented team. The Avengers handle big threats to Earth, the Defenders handle mob organizations that are too big for them to deal with on their own.The strike force response team is practically the original concept of what The Defenders were: a non-team who only teamed up when the situation arose but didn't spend the rest of the time together.
I remember starting up a thread shortly after the Avengers movie came out and questioning whether The Avengers really were a team or more of a response team. I recall everyone ridiculing that idea and saying "of course they are a team."
But the reality is that they aren't really a team and my question wasn't unreasonable at all. We can see this especially in all the Phase 2 movies. Iron Man hasn't hung out with his "team mates" in IM3. Thor is doing his own thing, and Cap has joined SHIELD and still serving with them two years on from the Avengers.
It seems then that there is at least a 2 year (probably more) period before the Avengers actually assemble again since the battle of New York.
If the FF were in the MCU, they would be a close-knit team who do hang out together, making them markedly different from the Avengers.
As for the TV Defenders, I don't know if they will also be essentially a response team who only come together when absolutely necessary. I don't really see DD hanging out with the others, but I could envision Cage and Fist forming a partnership, and Jessica Jones following.
In fact, I think it will be pretty close to what Daredevil tried to get together in the Ultimate comics. To bring down Kingpin, Daredevil tried to put together a team consisting of himself, Moon Knight, Spider-Man, and Ronin. I don't think he referred to them as the Defenders but I get the feeling that is what this team would basically be. A more selfless version of Heroes for Hire.
