The fact that Curt Connors and Eddie Brock are totally different characters would mean nothing - it just comes down to the fact that Lizard and Venom would be too much alike in action onscreen.

lizard is quite different from both spidey and venom, IMO

Doc Ock had wall-climbing ability and super-strength in the mechanical arms, but his body was normal.
Lizard and Venom do not use mechanical weapons, and they both have natural speed and agility like Spidey. They both climb walls in a way similar to Spidey, and they both use their teeth and claws when battling Spidey. So with the exception of the Lizard's tail and Venom's webbing/tentacles, the action scenes with Lizard and Venom would be similar.
Anyway - they're both monsters. There would not be two monsters in SM3.

Venom and Lizard are hulking teethy drooling monsters that are after Spider-Man. Plus the Lizard dwells in the sewers and comes out at night. Venom would most likely be out at night as well. Just too many similarities.

Especially since they seem to be going with scales like honeycombs for the symbiote suit. Maybe it will have some sort of reptilian DNA...to make Venom have big teeth and a long tongue. That would work perfect for Lizard and Connors set up.

When I think of the Lizard, I think of Darkman.
After the accident in the lab, he tries so desperately to be human again, and he will do anything it takes to achieve this. It's very tragic, but I love the idea of the Lizard in SM4.

Peter will also have to stop the Lizard's diabolical plan to turn humans into what he has become.
