It is ambiguous, but my theory is there was people developing powers naturally, but couldn't control them or they caused them pain. So they volunteered to these experiments by Strucker and co. The rest died but the twins survived. The fact that the twins were the only survivors indicates that there is something unique about their genetics.
I don't think the sceptre was used with the twins or other volunteers. It's just an artefact Strucker has recovered
